Fast Analysis of Vitamin D from Serum or Plasma
Dennis Nagtalon, Agilent Technologies
The analysis of vitamin D in a clinical research environment requires a quick and simple analytical method for rapid sample turnaround. LC–MS is ideal due to its fast analysis time and high-throughput capabilities with the ability to quantify both vitamin D forms. Using the 25-OH Vitamin D kit from ChromSystems GmbH (Munich, Germany), a 5-min method was developed providing simple, accurate, and rapid analysis of 25-OH vitamin D2 and D3 in plasma and serum. |

Simultaneous Determination of Therapeutic Drug Analytes in Human Plasma Using LC–MS-MS
Srividya Kailasam, Agilent Technologies
Simultaneous determination of atenolol, amlodipine, clonidine, enalapril, lisinopril, furosemide, hydrochlorothiazide, losartan, valsartan, telmisartan, and canrenone (the active metabolite of spironolactone) in human plasma by LC–MS-MS has been described in this application note. A simple protein precipitation sample preparation technique was used to extract the 11 drug analytes from plasma. Excellent linear calibration curves for all the target analytes were achieved with reproducibility. |

Analysis of Testosterone in Serum using LC–Triple Quadrupole MS-MS
Andre Szczesniewski, Agilent Technologies
A rapid method for the quantification of total testosterone in serum was developed using the Agilent 1200 Series Rapid Resolution LC (RRLC) system with the Agilent 6430 Triple Quadrupole LC–MS in atmospheric pressure chemical ionization (APCI) mode. A two dimensional LC approach was used to minimize matrix interferences and ion suppression, while reducing the analysis time 4.5 min. |

Simultaneous Analysis of Amino Acids and Acylcarnitines in Dried Blood Spots
David Presser, Agilent Technologies
A rapid method for the simultaneous detection and quantification of 37 acylcarnitines and 12 amino acids in dried blood spots was developed using the Agilent Triple Quadrupole LC–MS System in neutral loss scan, precursor ion scan, and multiple reaction monitoring (MRM) data acquisition modes. With a run time of 1.6 min, the analysis provides the ability to analyze 400–500 samples per day, and custom reports provide all the information needed to rapidly list the critical elements for the analysis. |

High Sensitivity SISCAPA-Based Peptide Quantitation Using UHPLC and the 6490 QQQ with iFunnel Technology
Dr. Leigh Anderson, SISCAPA Assay Technologies
This application note demonstrates the use of an integrated, automated, high-throughput MS-based peptide quantitation strategy to achieve sub-ng/mL detection levels of a target peptide following enrichment with SISCAPA® technology. Here we evaluate the sensitivity and throughput that can be achieved using more robust standard flow chromatography interfaced with the Agilent 6490 Triple Quadrupole MS with iFunnel technology to analyze peptides captured by a multiplex SISCAPA panel incorporating 11 rabbit monoclonal antibodies to specific peptides from 10 proteins spanning a wide concentration range in plasma. |
