Here we present an automated workflow for complete structural elucidation of glycans by combining MSn fragmentation and data processing. The combination of permethylation, MSn, and SimGlycan software enabled successful identification and differentiation of various structural isomers of chicken ovalbumin released glycans. The simplified data analysis time was reduced to a matter of minutes, thus enabling truly automated, high-throughput data analysis. |

Analysis of polycyclic aromatic hydrocarbons from vehicle exhaust using TD–GC/TOF MS

This Application Note describes how the combination of thermal desorption (TD) with gas chromatography (GC) and time-of-flight mass spectrometry (TOF MS), followed by data analysis using TargetView™ compound identification software, can be used to accurately identify trace-level polycyclic aromatic hydrocarbons (PAHs) in a complex vehicle exhaust particulate sample. The sensitivity and ‘classical’ spectra produced by BenchTOF-dx allow accurate identification of PAHs with near-identical fragmentation patterns, while TargetView enables GC profiles of trace-level analytes to be visualised even when hidden under much more abundant components. |

Automated Solid Phase Extraction of Semi-Volatile Organic Compounds (AB 8270 SIM) in Water

EPA Method 8270 is used to determine the concentration of semi-volatile organic compounds in extracts prepared from solid waste matrices such as soils, air sampling media and water samples. Extractions from water samples can be particularly difficult and time consuming due to the problematic analytes that need to be extracted, concentrated and analyzed as well as the large quantities of solvent the manual sample prep process requires. Laboratories are required to report analytes at lower reporting limits than previously required, often needing to employ SIM scan variations of the method. The following application outlines the complete automation of a modified 8270 extraction, delivering a complete, out-of-the box MDL study demonstrating both precision and accuracy for mixed classes of low-level analytes.

Extraction of Cortisol From Human Saliva Using ISOLUTE®SLE+ Plates Prior to LC-MS/MS Analysis

Cortisol is a steroid hormone that when measured from saliva can be used as an indication of stress. This methodology has been designed to give an effective and efficient supported liquid extraction protocol for the clean-up and concentration of salivary cortisol levels. Analyte recoveries achieved using this method ranged from 96-99% with RSDs below 3% (n=7) for all analytes. |

Determination of Phthalates Leached from Toys into Artificial Saliva

Phthalates are plasticizers added to polymers to make them softer and more flexible. They are commonly used in the manufacturing of toys. Phthalates are not permanently bonded in polymers and they may be released through touching, licking, or chewing. This new method artificially reproduces the licking and chewing actions of children by shaking cut toys in artificial saliva. Following extraction, phthalate levels are determined by GC/MS. |

EPA 8330A Analysis of Explosives Using Agilent Poroshell 120 EC-CN and EC-C18 Columns

A group of 14 nitroaromatics and nitramines was analyzed according to EPA Method 8330A for the trace analysis of explosive residues by HPLC/UV. The analysis was improved by using superficially porous Agilent Poroshell 120 columns as compared to the traditional 5 μm columns suggested by the United States Environmental Protection Agency (EPA). Two complimentary phases were used; Poroshell 120 EC-CN and Poroshell 120 EC-C18. As recommended by the EPA, the Poroshell 120 EC-CN column was a good confirmation column when paired with a Poroshell 120 EC-C18 column due to its orthogonal selectivity. |

When the fluorides, chlorides, and sulfur in the butane react, they can produce extremely corrosive acids, which can cause gas leaks. USP Analysis of Warfarin Sodium Tablets Using Agilent Poroshell 120 EC-CN and SB-C8 Columns

Warfarin, warfarin-related compound A, and propylparaben were analyzed according to the United States Pharmacopeia (USP) analysis for warfarin sodium tablets. The chromatographic purity and assay analyses were improved by using superficially porous Agilent Poroshell 120 columns as compared to the USP-suggested 5 μm columns. Each method was adjusted within the guidelines in USP Chapter 621 toallow for time and solvent savings with the Poroshell 120 columns. All chromatographic system requirements were met with the improved superficially porous column analyses. |
