SEC-Light Scattering for Antibody Drug Conjugate Characterization
Wyatt Technology
Among the factors critical to effective ADC design is the drug antibody ratio (DAR). The DAR describes the degree of drug addition, which directly impacts both potency and potential toxicity of the therapeutic, and can have significant effects on properties such as stability and aggregation. Determination of DAR is, therefore, of critical importance in the development of novel ADC therapeutics. We present here a method for DAR determination based on SEC-MALS in conjunction with UV absorption and differential refractive index detection. Read More |

Analysis of Membrane Protein by Multi-Detector SEC
Malvern Instruments
Detailed description of how a protein detergent complex (PDC), formed during the purification of a bacterial membrane protein involved in multidrug resistance, can be fully characterized. A triple detection SEC system, equipped with light scattering and viscometer detectors, was used to measure the molecular weight and composition of the PDC and of the free detergent micelles. Validation of PAHs in Soil and Sediment Samples Using Pressurized Liquid Extraction, Cleanup, and Concentration
Phillip Bassignani, FMS,Inc.
PAHs are a group of organic compounds consisting of two or more benzene rings and are often the byproduct of petroleum combustion. Due to their carcinogenic characteristics at relatively low concentra-tions, they are of particular environmental concern. Seven PAHs have been classified by the US EPA as probable human carcinogens and their toxic characteristics and persistent nature place them among the most extensively monitored organic contaminants. The following procedure details the use of pressurized liquid extraction (PLE) in conjunction with automated silica gel cleanup (PowerPrep) to deliver a robust, efficient sample prep process for soils and sediments designated for PAH analysis. Outlined, are an initial precision and recovery study (IPR), a matrix detection level study (MDL), and a matrix validation of a NIST reference material. Read More |

Metabolic Profiling of Coffee
Dr. Verena Tellstrom, Dr. Alexander Harder, Klaus Meyer and Dr. Aiko Barsch, Bruker Daltonics
With worldwide consumption of coffee at 4.5 million tons, quality control is an important issue and can be established on a large scale through metabolomic profiling. In this study, 13 types of coffee capsule extracts were analyzed to correlate high resolution LC-MS data to the manufactuturer's description via a non-targeted metabolomics approach. Read More |

Troubleshooting System Problems Using Waters Neutrals Quality Control Reference Materials
Kenneth D. Berthelette, Mia Summers, and Kenneth J. Fountain, Waters Corporation
In this application, six common chromatographic problems are examined to demonstrate the utility of the Neutrals QCRM in rapidly diagnosing problems on an HPLC or UPLC system. After repairs were made, the Neutrals QCRM was used to quickly confirm that the system was back to operating optimally. By using the Neutrals QCRM to check system functionality, data quality can be assured and the system can be used with confidence. Read More |
