Characterization of Glycosylation in the Fc Region of Therapeutic Recombinant Monoclonal Antibody

In this note, we describe a fast and efficient liquid chromatography time-of-flight mass spectrometry approach to characterize an IgG derived from CHO cells.  An Agilent Poroshell 300SB-C3, 5 µm superficially porous IgG1 column was used to obtain accurate glycoform masses of an intact IgG1, with a subsequent analysis of papain-digested IgG1 to obtain site-specific glycosylation profile information of the Fc region for further characterization of IgG1 heterogeneity.  Additionally, a subsequent mAb-glyco chip LC–MS analysis was used to build a glycan accurate mass database for additional validation of the glycans identified in the TOF mass analysis. Providing a Universal, One-step Alternative to Liquid-Liquid Extraction in Bioanalysis

The Ostro sample preparation plate and a generic liquid-liquid extraction (LLE) method were compared for the extraction of a diverse group of pharmaceutical drugs. With no method development, analyte recovery was high, >75%, for polar and nonpolar, acidic, and basic analytes alike, and 95% of phospholipids were removed relative to LLE. Ostro extraction of 96 samples was achieved in half the time relative to 96-well LLE and in less than 1/10th the time if LLE was performed in individual tubes. Read More
