Solvent Selection for Flash Purification with the 971-FP: The "Guide Me" Feature

IntelliFlash™ software used with the 971-FP flash purification instrument offers a "Guide Me" feature, which simplifies the decision processes needed to define the optimum purification method. The stages of method definition and the information needed for each step are all contained within this intuitive set-up wizard. This note examines the effect of solvent choice on the separation of a standard test mix containing ethyl benzene, ethyl benzoate and acetophenone.

Analysis of Peptides on a PLRP-S 100Å 10 µm with ELS Detection and Acetonitrile-free Eluents

Due to the current global acetonitrile shortage, Varian has been investigating a range of alternative solvents that could be used for reversed-phase HPLC analysis of peptides, which maintain the efficiency and selectivity that a PLRP-S 100Å 10 µm gives for this application when used with acetonitrile. One of the disadvantages of changing from acetonitrile to an alternative solvent is the increased background absorbance of the mobile phase eluents when run on a UV detector. Many solvents, such as methanol, ethanol, and iso-propanol, absorb UV strongly in this region giving baseline drift and noise that interferes with the separation, therefore this report details how a Varian 385-LC ELS detector can be used to prevent this.

An Empirical Evaluation of HILIC and Monolithic Columns for Supercritical Fluid Chromatography (SFC) Applications

A column comparison was conducted on a Spherisorb® silica column, X-BridgeTM HILIC column and OnyxTM monolithic column for the separation of polar compounds in supercritical fluid chromatography (SFC). A mixture of 14 compounds was run under gradient and isocratic conditions on all three columns. The results indicate that both the X-BridgeTM HILIC and the OnyxTM monolithic columns have great potential for use in SFC applications.
