Featured Application Note:
Quantification of EPA 1694 Pharmaceuticals and Personal Care Products in Water at the ng/L Level Utilizing Online Sample Preparation with LC-MS/MS
By Kevin McHale, Mark Sanders, Thermo Fisher Scientific

The goal of this application note is to demonstrate a progressive approach to analyzing PPCPs in environmental sources of water at the ng/L level with online sample preparation using small volumes of water, thus saving time and reducing the cost of analysis.

Hyphenated
Characterization of monoclonal antibodies by Size Exclusion Chromatography
Srividya Kailasam, Jochen Strassner, Faizy Ahmed and Martin Vollmer Agilent Technologies Bangalore, India and Waldbronn, Germany, Agilent Technologies

Purity and stability of two monoclonal antibodies (mAb) preparations were investigated using the Agilent 1260 Bio-inert LC System in combination with the Agilent BioSEC columns. Superior Resolution of variants from the main antibody peak was achieved and compared to the results obtained with a competitive column.

Chiral LC/MS/MS Method Development of Stereoisomeric Pharmaceutical Compounds
Philip J. Koerner, Kari Carlson, Liming Peng, Swapna Jayapalan, Tivadar Farkas, Phenomenex

Five different Lux® polysaccharide-based chiral stationary phases were explored in the reversed phase elution mode using mobile phases consisting of 0.1 % formic acid in acetonitrile or methanol to demonstrate the feasibility of LC/MS/MS analysis of a variety of acidic pharmaceutical racemates.

209 PCB-Congeners using LECO Pegasus GCxGC-TOFMS
Cory Fix and Mark Libardoni, LECO

The analysis of 209 individual PCB congeners using the LECO Pegasus 4D GCxGC-TOFMS instrument with ChromaTOF software is described here. Retention times in the 1st and 2nd dimensions were recorded for all 209 PCB congeners and are listed (including all critical pairs). Separation and identification of critical pairs was performed primarily in the 2nd dimension, highlighting the power of the comprehensive LECO Pegasus 4D GCxGC-TOFMS technique.
Fast GC-MS/MS for High Throughput Pesticides Analysis
Igor Fochi, Elena Ciceri, Hans-Joachim Huebschmann Ciro D’Aniello, Thermo Fisher Scientific

This application describes a Fast GC-MS method for high sample throughput and improved analytical results. The described Fast GC-MS approach delivers matrix robustness with reliable integration of short Fast GC peaks with high selectivity and sensitivity.

Sample Prep
Heparin analysis and characterization
Jams Knobloch and Patrick Shaklee, Wyatt Technology Corporation

The U.S. Food and Drug Administration (FDA) has scrutinized LMW-heparin products closely due to their inherent variability and polydispersity. Previously, these materials had to be characterized by relative techniques involving viscometry or standard elution-based chromatographic methods.

Predicting Drug Metabolism using Electrochemistry/MS
A, Kraj, H-J. Brouwer, M. Eysberg, Antec

For the prediction and mimicking of drug/xenobiotic metabolism Antec has developed a dedicated Electrochemistry system, called ROXY™ EC. The system can be used up-front MS and allows to generate and screen for potential oxidative metabolites, mimicking P450 enzymatic reactions in a few minutes.

Determination of Polyphosphates In Food Products
Weerapong Worawirunwong, Dionex Corporation

Polyphosphates are sometimes added to food products as a preservative (e.g. fresh fish) but the amount that can be added is limited in some countries. This application update shows the determination of polyphosphates in a food sample using a hydroxide selective column and continuous suppression of a strong eluent.

Determination of Phosphorous by Ion Chromatography
Rolf Maurer & Bern Sheldon, Dionex Corporation

A heart-cutting technique is used to take a small amount of phosphate from the separation of the sample that contains a large amount of sulfate and placed on a concentrator. After the other ions are eluted from the column and the column is reequilbrated, the phosphate from the concentrator is placed back on the same column for separation and quantification.

Dioxin in Drinking Water by One Step Solid Phase Extraction
Phillip Bassignani, FMS, Inc

Dioxins are a group of polyhalogenated compounds which occur as by-products in the manufacture of organochlorides, in the incineration of chlorine-containing substances such as PVC (polyvinyl chloride), in the bleaching of paper, and from natural sources such as volcanoes and forest fires. As known carcinogens, dioxins and dioxin like compounds have been linked to developmental abnormalities, nervous system, pathology, diabetes, thyroid disorders and damage to the immune system. For these reasons, it is becoming increasingly important to determine dioxin free drinking water and food sources. This application note demonstrates the use of the FMS automated One Step SPE system in water samples.

Simplified SPE Approach for SAMHSA’s Low Cutoff Drugs of Abuse Panel
Michael Rummel, Matthew Trass, Seyed Sadjadi, Erica Pike, Phenomenex

Utilizing a single Solid Phase Extraction (SPE), Strata™-X-Drug B, sorbent and only 3 extraction methods, 11 drugs from 6 different Substance Abuse and Mental Health Services Administration (SAMHSA) regulated drug classes were effectively extracted and analyzed by LC/MS and GC/MS. The extraction resulted in consistent, high recoveries of each drug analyzed that conformed to the new lower cutoff levels that SAMHSA has recently enforced.
