Fast DPX-LC/MS/MS Drug Analysis.
Rapid Disposable Pipette Extraction (DPX) of drugs from biological fluids is presented. DPX is shown to remove matrix interference, reducing ion suppression and solvent consumption and leading to improved limits of determination. Automated DPX extraction is coupled with sample introduction to the LC/MS/MS system.

Preparative Scale Purification of Bradykinin by Volume Overload

Where a peptide is a biopharmaceutical API candidate the amount required will increase as it moves through clinical trials to product. When developing purification methods the required “product” API, quantity and purity, should be considered. Bradykinin, a physiologically and pharmacologically active peptide of the kinin family is used in the development of antagonists and therapies for hereditary angioedema. In this application note, preparative scale purification of a sample of synthetic bradykinin using a volume overload regime is described.

Synthetic peptide production is one of the major growth areas in biopharmaceutical drug development. Many of the processes used for synthetic peptide purification are based around small particle high performance RP-silica materials with gradient elution and so have high capital and running costs. This application note discusses the possibility of using a larger robust polymeric particle, which would be expected to have a longer column lifetime under aggressive conditions, and would offer additional savings in the cost of materials required for preparative scale purification.
