One of the most difficult challenges confronting chromatographers is the retention and separation of highly polar water-soluble organic compounds. Unfortunately, many chromatographers do not understand the reasons for poor polar compound retention.  Presented in this seminar is a practical explanation of stationary phase chemistry and its role in polar compound retention.  Building upon this basic understanding, a description is provided that explains how a stationary phase can provide enhanced polar compound retention and longer column lifetimes in reversed-phase separations.  Finally, we will learn about how particle substrate and stationary phase research can provide similar performance benefits in Ultra Performance LC separations.

Specific topics include:

  • Chromatographic techniques that have been used to retain, separate and detect polar, water-soluble organic compounds
  • What hydrophobic collapse really is (and more importantly, what it is not)
  • An explanation of why embedded polar group stationary phases actually cause less retention for polar compounds in reversed-phase HPLC
  • How an intelligently designed C18 stationary phase can retain polar compounds while thriving in acidic, highly aqueous mobile phases
  • Examples of how bonded phase research along with particle morphology manipulation can offer improved UPLC and UHPLC separations
