Given the heterogeneity and complexity of biologics, including monoclonal antibodies, advance analytical techniques are now essential for characterization, identification, and quantitation. Reversed phase liquid chromatography with mass spectrometry (RP‑MS) is the primary tool for confirmation of intact mass and characterization of post translational modifications (PTMs), such as glycosylation, oxidation, and deamidation. The use of MS detection adds another dimension to method development, as not only is it essential that the method has the capability of resolving the critical pairs, but it must also be accomplished using mobile phases and columns that are compatible with the detector.
